Finally, the cellular uptake of curcumin and curcumin NCs was evaluated on Olfactory Ensheathing Cells (OECs). Our results showed that the OECs efficiently took up the NCs compared to the free curcumin, showing that NCs can ameliorate drug permeability.During the course of our search for novel biologically active metabolites from tropical fungi, we are using chemotaxonomic and taxonomic methodology for the preselection of interesting materials. Recently, three previously undescribed benzo[j]fluoranthenes (1-3) together with the known derivatives truncatones A and C (4, 5) were isolated from the stromata of the recently described species Annulohypoxylon viridistratum collected in Thailand. SGC 0946 purchase Their chemical structures were elucidated by means of spectral methods, including nuclear magnetic resonance (NMR) spectroscopy and high-resolution mass spectrometry (HR-MS). The new compounds, for which we propose the trivial names viridistratins A-C, exhibited weak-to-moderate antimicrobial and cytotoxic activities in cell-based assays.Neural stem cells are fundamental to development of the central nervous system (CNS)-as well as its plasticity and regeneration-and represent a potential tool for neuro transplantation therapy and research. This study is focused on examination of the proliferation dynamic and fate of embryonic neural stem cells (eNSCs) under differentiating conditions. In this work, we analyzed eNSCs differentiating alone and in the presence of sonic hedgehog (SHH) or triiodothyronine (T3) which play an important role in the development of the CNS. We found that inhibition of the SHH pathway and activation of the T3 pathway increased cellular health and survival of differentiating eNSCs. In addition, T3 was able to increase the expression of the gene for the receptor smoothened (Smo), which is part of the SHH signaling cascade, while SHH increased the expression of the T3 receptor beta gene (Thrb). This might be the reason why the combination of SHH and T3 increased the expression of the thyroxine 5-deiodinase type III gene (Dio3), which inhibits T3 activity, which in turn affects cellular health and proliferation activity of eNSCs.Several studies show that triple-negative breast cancer (TNBC) patients have the lowest vitamin D concentration among all breast cancer types, suggesting that this vitamin may induce a protective effect against TNBC. This effect of the active metabolite of vitamin D, 1α,25-dihydroxyvitamin D3 (1,25(OH)2D), can be attributed to its potential to modulate proliferation, differentiation, apoptosis, inflammation, angiogenesis, invasion and metastasis and is supported by many in vitro and animal studies, but its exact mechanism is poorly known. In a fraction of TNBCs that harbor mutations that cause the loss of function of the DNA repair-associated breast cancer type 1 susceptibility (BRCA1) gene, 1,25(OH)2D may induce protective effects by activating its receptor and inactivating cathepsin L-mediated degradation of tumor protein P53 binding protein 1 (TP53BP1), preventing deficiency in DNA double-strand break repair and contributing to genome stability. Similar effects can be induced by the interaction of 1,25(OH)2D with proteins of the growth arrest and DNA damage-inducible 45 (GADD45) family. Further studies on TNBC cell lines with exact molecular characteristics and clinical trials with well-defined cases are needed to determine the mechanism of action of vitamin D in TNBC to assess its preventive and therapeutic potential.Metal-based additive manufacturing (AM) is a disruptive technique with great potential across multiple industries; however, its manufacturing quality is unstable, leading to an urgent requirement for component properties detection. The distribution of grain size has an important effect on many mechanical properties in AM, while the distribution of added elements, such as titanium (Ti), has a measurable effect on the grain size of an aluminum (Al) alloy. Therefore, the detection of the distributions of grain size and elements is of great significance for AM. In this study, we investigated the distribution of grain size and elements simultaneously for wire + arc additive manufacturing (WAAM) with an Al alloy using laser opto-ultrasonic dual (LOUD) detection. The average grain size obtained from the acoustic attenuation of ultrasonic signals was consistent with the results of electron backscatter diffraction (EBSD), with a coefficient of determination (R2) of 0.981 for linear fitting. The Ti element distribution obtained from optical spectra showed that the enrichment of Ti corresponded to the grain refinement area in the detected area. The X-ray diffraction (XRD) spectra showed that the spectral peaks were moved from Al to AlTi and Al2Ti forms in the Ti-rich areas, which confirmed the LOUD results. The results indicated that LOUD detection holds promise for becoming an effective method of analyzing the mechanical and chemical properties of components simultaneously, which could help explain the complex physical and chemical changes in AM and ultimately improve the manufacturing quality.Porous titania was successfully synthesized by an ultrasound-assisted sol-gel route. The synthesis process was empirically modeled and optimized using the response surface methodology (RSM). Input variables adopted for optimization dealt with the weight ratio of precursors (r) and the sonication time (t), representing the used factors in the synthesis procedure. With regard to application, the synthesized TiO2 samples were tested for the photodegradation of two water-soluble organic pollutants under UV-Vis irradiation. Optimal conditions for the efficient pollutants’ photodegradation were found to involve a precursors ratio of 3 and a sonication time of 60 min. Thus, the M5 sample prepared under the founded optimal conditions yielded the maximal removal efficiencies of 98.4% and 46.3% for the photodegradation of CR dye and 2,4-D herbicide, respectively. In addition, the photodegradation kinetics revealed the pseudo first-order rate constants, showing the photodegradation of CR (k1 = 8.86 × 10-2 min-1) by M5 sample is about 1.
