Evidence of the involvement of epigenetics in pathologies such as cancer, diabetes, and neurodegeneration has increased global interest in epigenetic modifications. For nearly thirty years, it has been known that cancer cells exhibit abnormal DNA methylation patterns. In contrast, the large-scale analysis of histone post-translational modifications (hPTMs) has lagged behind because classically, histone modification analysis has relied on site specific antibody-based techniques. Mass spectrometry (MS) is a technique that holds the promise to picture the histone code comprehensively in a single experiment. Therefore, we developed an MS-based method that is capable of tracking all possible hPTMs in an untargeted approach. In this way, trends in single and combinatorial hPTMs can be reported and enable prediction of the epigenetic toxicity of compounds. Moreover, this method is based on the use of human cells to provide preliminary data, thereby omitting the need to sacrifice laboratory animals. Improving the workflow and the user-friendliness in order to become a high throughput, easily applicable, toxicological screening assay is an ongoing effort. At the Berkeley Space Sciences Laboratory, we are working to further expand this technology by developing analytical instruments to chemically explore our solar system. This effort requires improving the quality and reliability of glass microfabrication through quality control procedures at every stage of design and manufacture. This manuscript provides detailed information on microfabrication technology for the production of high-quality glass microfluidic chips in compliance with industrial standards and space flight instrumentation quality control.•The methodological protocol provided in this paper includes the scope of each step of the manufacturing process, materials and technologies recommended and the specific challenges that often confront the process developer.•Types and sources of fabrication error at every stage have been identified and their solutions have been proposed and verified.•We present robust and rigorous manufacturing and quality control procedures that will assist other researchers in achieving the highest possible quality glass microdevices using the latest apparatus in a routine and reliable fashion.Gastrointestinal stromal tumors are rare, and in pregnancy they are extremely rare. We present a case of a maternal gastrointestinal stromal tumor found in the second trimester of pregnancy. A 29-year-old woman, gravida 1 para 0, complained of bloody vomiting at 14 weeks of gestation. She had no significant medical history. We performed plain computed tomography and upper gastrointestinal endoscopy. Precise examination revealed a large mass in the stomach and an exposed blood vessel on the surface. An exposed blood vessel can be harmful for mother and fetus as it might rupture during the pregnancy. We performed a distal gastrectomy at 16 weeks of gestation. Histology confirmed a localized gastrointestinal stromal tumor with a high risk of recurrence, and adjuvant imatinib was recommended. The patient elected to delay adjuvant imatinib until after delivery. The postoperative and antenatal course was favorable, and the patient was followed up by ultrasound every 2 months after the operation. After she gave birth at 40 weeks of gestation, she started adjuvant imatinib 400 mg/day. There was no evidence of recurrence 1 year after surgery. There are no guidelines for the management of gastrointestinal stromal tumors in pregnancy. Given the treatment challenges, we believe that pregnant patients should be managed by a multidisciplinary team with expertise in gastrointestinal tumors and fetal-maternal medicine.Nanocomposite materials are emerging in popularity due to their enhanced performance over the constituent materials. In this work, we report the fabrication of zinc oxide cobalt oxide nanocomposites in a simple, fast and room temperature synthesis with good productivity. The nanocomposites synthesized were characterized by SEM, XPS and UV-Visible spectroscopy to analyze their morphology, composition, chemical states, optical absorption, band gap etc. The nanocolloids of the composite were drop casted to form thin films for photocatalytic studies. In SEM analysis, the morphological transformation of the material is observed where it transformed from agglomerated spherical particles to petals shaped and then to partially spherical forms due to pulsed laser irradiation. XPS analysis showed a gradual change in oxygen high resolution spectra in the samples with respect to the concentration difference of cobalt oxide. Proteases inhibitor The optical studies show an enhanced absorption in visible region for the nanocomposite and the energy band gap reduced to 2.4 eV. All the thin films of nanocomposite showed photocatalytic decay of methylene blue dye under visible light irradiation. The results of this study support the effective use of laser irradiation in liquid to obtain nanocomposites of metal oxides for photocatalytic applications.
This study presents a protocol for a mixed method study on the development and evaluation of the effectiveness of mobile phone-based intervention for enhancing career competencies of managers in the Malaysian public service. Career competencies, being central to the self-management and development of one’s career, are necessary for proactive management of careers; they help to guide the individuals towards subjective career success which subsequently leads to organization success. However, there is a lack of career competencies intervention practices in the Malaysian public service. Therefore, this study aims to develop and evaluate the effectiveness of a user and expert centric mobile phone-based career intervention program of career competencies among Malaysian public managers. In this study, the Social Cognitive Career Theory (SCCT) is adopted as the framework to develop mobile phone-based intervention for career competencies.
This is a sequential exploratory mixed method study which involves, in Phase 1, a qualitative approach reviewing of literature and interviewing experts and public servants who are users of mobile phones so as to obtain their perspectives regarding the components in the career intervention module and mobile phone application features.
