The role of the gut microbiome in human health is becoming apparent. The major functional impact of the gut microbiome is transmitted through the microbial metabolites that are produced in the gut and interact with host cells either in the local gut environment or are absorbed into circulation to impact distant cells/organs. Short-chain fatty acids (SCFAs) are the major microbial metabolites that are produced in the gut through the fermentation of non-digestible fibers. SCFAs are known to function through various mechanisms, however, their signaling through free fatty acid receptors 2 and 3 (FFAR2/3; type of G-coupled protein receptors) is a new therapeutic approach. FFAR2/3 are widely expressed in diverse cell types in human and mice, and function as sensors of SCFAs to change several physiological and cellular functions. FFAR2/3 modulate neurological signaling, energy metabolism, intestinal cellular homeostasis, immune response, and hormone synthesis. FFAR2/3 function through Gi and/or Gq signaling, that is mediated through specific structural features of SCFAs-FFAR2/3 bindings and modulating specific signaling pathway. In this review, we discuss the wide-spread expression and structural homologies between human and mice FFAR2/3, and their role in different human health conditions. This information can unlock opportunities to weigh the potential of FFAR2/3 as a drug target to prevent human diseases.Glucocorticoids are frequently used as anti-inflammatory and immunosuppressive agents. However, high doses and/or prolonged use induce undesired secondary effects such as muscular atrophy. Recently, de novo expression of connexin43 and connexin45 hemichannels (Cx43 HCs and Cx45 HCs, respectively) has been proposed to play a critical role in the mechanism underlying myofiber atrophy induced by dexamethasone (Dex a synthetic glucocorticoid), but their involvement in specific muscle changes promoted by Dex remains poorly understood. Moreover, treatments that could prevent the undesired effects of glucocorticoids on skeletal muscles remain unknown. In the present work, a 7-day Dex treatment in adult mice was found to induce weight loss and skeletal muscle changes including expression of functional Cx43/Cx45 HCs, elevated atrogin immunoreactivity, atrophy, oxidative stress and mitochondrial dysfunction. All these undesired effects were absent in muscles of mice simultaneously treated with Dex and vitamin E (VitE). Moreover, VitE was found to rapidly inhibit the activity of Cx HCs in freshly isolated myofibers of Dex treated mice. Exposure to alkaline pH induced free radical generation only in HeLa cells expressing Cx43 or Cx45 where Ca2+ was present in the extracellular milieu, response that was prevented by VitE. Besides, VitE and two other anti-oxidant compounds, Tempol and Resveratrol, were found to inhibit Cx43 HCs in HeLa cells transfectants. click here Thus, we propose that in addition to their intrinsic anti-oxidant potency, some antioxidants could be used to reduce expression and/or opening of Cx HCs and consequently reduce the undesired effect of glucocorticoids on skeletal muscles.(1) Background Ranking traits are used commonly for breeding purposes in several equine populations; however, implementation is complex, because the position of a horse in a competition event is discontinuous and is influenced by the performance of its competitors. One approach to overcoming these limitations is to assume an underlying Gaussian liability that represents a horse’s performance and dictates the observed classification in a competition event. That approach can be implemented using Montecarlo Markov Chain (McMC) techniques with a procedure known as the Thurstonian model. (2) Methods We have developed software (GIBBSTHUR) that analyses ranking traits along with other continuous or threshold traits. The software implements a Gibbs Sampler scheme with a data-augmentation step for the liability of the ranking traits and provides estimates of the variance and covariance components and predictions of the breeding values and the average performance of the competitors in competition events. (3) Results The results of a simple example are presented, in which it is shown that the procedure can recover the simulated variance and covariance components. In addition, the correlation between the simulated and predicted breeding values and between the estimates of the event effects and the average additive genetic effect of the competitors demonstrates the ability of the software to produce useful predictions for breeding purposes. (4) Conclusions the GIBBSTHUR software provides a useful tool for the breeding evaluation of ranking traits in horses and is freely available in a public repository (https//github.com/lvaronaunizar/Gibbsthur).Multifunctional metasurfaces play an important role in the development of integrated optical paths. However, some of the realizations of current multifunctional metasurface devices depend on polarization selectivity, and others change the polarization state of the outgoing light. Here, based on vanadium dioxide (VO2) phase change material, a strategy to design a meta-duplex-lens (MDL) is proposed and numerical simulation calculations demonstrate that at low temperature (about 300 K), VO2 behaves as a dielectric so that the MDL can act as a transmission lens (transmission efficiency of 87.6%). Conversely, when VO2 enters the metallic state (about 355 K), the MDL has the ability to reflect and polymerize electromagnetic waves and works as a reflection lens (reflection efficiency of 85.1%). The dielectric waveguide and gap-surface plasmon (GSP) theories are used in transmission and reflection directions, respectively. In order to satisfy the coverage of the phase gradient in the range of 2π in both cases, we set the antenna as a nanopillar with a high aspect ratio. It is notable that, via symmetrical antennas acting in concert with VO2 phase change material, the polarization states of both the incident light and the outgoing light are not changed. This reversible tuning will play a significant role in the fields of imaging, optical storage devices, communication, sensors, etc.
